The Coordination Status of the Children\u27s Mental Health Services in Minnesota
The purpose of the present study is to describe what services children with severe emotional disturbance (SED) have received before acute care hospitalization. According to the Minnesota children\u27s Mental Health Act of 1989, children should be receiving a comprehensive set of services. This means that the children\u27s mental health system is to be integrated and coordinated with all child-serving systems at the state, local and individual levels . This study examines the following three research questions: a) Is coordination of services being provided across disciplines?, b) Are children receiving case-management services?, and c) Are children receiving services in the least restrictive setting first before moving to more restrictive environments?. Twenty (20) adolescents were interviewed on a psychiatric unit of an acute care hospital. The findings indicate that all services are being provided as described in the research questions and are in accordance with the 1989 Children\u27s Mental Health Act
Alternative Service Delivery Models for Students with Learning Disabilities
More than 40 years of intervention research describes effective interventions for increasing the academic performance of students with learning disabilities. However, the performance and outcomes for students with learning disabilities remain discouraging, especially in light of the increasing cost to provide special education services. Between 60 â 70 percent of students with learning disabilities are unable to meet grade level performance standards in the basic academic areas of reading, writing and mathematics. The disconnect between research and practice suggests a need for rethinking special education service delivery. This paper reports the results of a program evaluation of a not-for-profit center specializing in working with students with learning disabilities, comparing the pre to post test gains and program costs to those typically seen in public education special education systems. A paired samples t-test examined pre to post test differences in reading, math and writing performance for a total of 104 students with learning disabilities between the ages of 6â17. Results were statistically and practically significant across all areas, and these results were obtained at costs much lower than those incurred under the current public education model. Implications for future research are discussed

Examining Interrater Agreement Analyses of a Pilot Special Education Observation Tool
This paper reports the results of interrater agreement analyses on a pilot special education teacher evaluation instrument, the Recognizing Effective Special Education Teachers (RESET) Observation Tool (OT). Using evidence-based instructional practices as the basis for the evaluation, the RESET OT is designed for the spectrum of different instructional needs found within special education classrooms. The RESET OT informs what Danielson (2011) maintains are the two features of a teacher evaluation system 1) ensuring teacher quality and 2) promoting professional development. In June 2012, six special education teachers participated in a data coding session using the pilot RESET OT to evaluate video observations of special education instructional practice from the 2011-2012 school year. The teacher coders received an introductory training session to the RESET OT, and participated in two whole-group coding sessions before completing individual coding assignments. The results of the interrater agreement analysis report weak to no agreement within specific instructional practices, indicating the need for 1) additional research and development on the RESET OT 2) repeating the data coding session using a group of teacher coders who have received in-depth training on the RESET OT and evidence-based instructional practices, and 3) further investigation into the specific components of evidence-based instructional practice and how these might be applied across settings
Validating an Observation Protocol to Measure Special Education Teacher Effectiveness
This study used Kaneâs (2013) Interpretation/Use Argument (IUA) to measure validity on the Recognizing Effective Special Education Teachers (RESET) observation tool. The RESET observation tool is designed to evaluate special education teacher effectiveness using evidence-based instructional practices as the basis for evaluation. In alignment with other studies (Bell et al., 2012), we applied and interpreted Kaneâs (2006) four inferences for trait observation: scoring, generalization, extrapolation, and decision rules. Results from this study show that acceptable levels of validity are promising for the RESET observation tool. Because the RESET observation tool is premised on the idea that by increasing the use of evidence-based practices, student achievement will also increase, further investigations into the relationship between fidelity of implementation of instruction and student achievement will be critical for moving project work forward
Differences in the Determinants of Retirement Preparation between Farm and Nonfarm Households
As principal farm operators age, retirement and succession planning has become increasingly important to the U.S. agriculture industry. This study examined differences in the determinants of retirement preparation between farm and nonfarm households using the Survey of Consumer Finances. Factors such as risk preferences, financial capability, human capital, and other demographic characteristics of farmers that may play a role in their decision to plan for retirement were examined. Retirement planning was investigated by running three separate sets of logistic regressions on the overall sample, farm households, and nonfarm households. Likelihood of consulting a financial planner, expecting to leave a bequest, and household net worth were used as dependent variables. Results indicate some significant differences between farm and nonfarm households and highlight limitations in currently available data sets for studies such as this one. Implications for practitioners, researchers, and policymakers regarding farm family retirement and succession planning are discussed
Helicobacter pylori Exploits a Unique Repertoire of Type IV Secretion System Components for Pilus Assembly at the Bacteria-Host Cell Interface
Colonization of the human stomach by Helicobacter pylori is an important risk factor for development of gastric cancer. The H. pylori cag pathogenicity island (cag PAI) encodes components of a type IV secretion system (T4SS) that translocates the bacterial oncoprotein CagA into gastric epithelial cells, and CagL is a specialized component of the cag T4SS that binds the host receptor α5ÎČ1 integrin. Here, we utilized a mass spectrometry-based approach to reveal co-purification of CagL, CagI (another integrin-binding protein), and CagH (a protein with weak sequence similarity to CagL). These three proteins are encoded by contiguous genes in the cag PAI, and are detectable on the bacterial surface. All three proteins are required for CagA translocation into host cells and H. pylori-induced IL-8 secretion by gastric epithelial cells; however, these proteins are not homologous to components of T4SSs in other bacterial species. Scanning electron microscopy analysis reveals that these proteins are involved in the formation of pili at the interface between H. pylori and gastric epithelial cells. ÎcagI and ÎcagL mutant strains fail to form pili, whereas a ÎcagH mutant strain exhibits a hyperpiliated phenotype and produces pili that are elongated and thickened compared to those of the wild-type strain. This suggests that pilus dimensions are regulated by CagH. A conserved C-terminal hexapeptide motif is present in CagH, CagI, and CagL. Deletion of these motifs results in abrogation of CagA translocation and IL-8 induction, and the C-terminal motifs of CagI and CagL are required for formation of pili. In summary, these results indicate that CagH, CagI, and CagL are components of a T4SS subassembly involved in pilus biogenesis, and highlight the important role played by unique constituents of the H. pylori cag T4SS
The path to triacylglyceride obesity in the sta6 strain of Chlamydomonas reinhardtii
When the sta6 (starch-null) strain of the green microalga Chlamydomonas reinhardtii is nitrogen starved in acetate and then âboostedâ after 2 days with additional acetate, the cells become âobeseâ after 8 days, with triacylglyceride (TAG)-filled lipid bodies filling their cytoplasm and chloroplasts. To assess the transcriptional correlates of this response, the sta6 strain and the starch-forming cw15 strain were subjected to RNA-Seq analysis during the 2 days prior and 2 days after the boost, and the data were compared with published reports using other strains and growth conditions. During the 2 h after the boost, âŒ425 genes are upregulated â„2-fold and âŒ875 genes are downregulated â„2-fold in each strain. Expression of a small subset of âsensitiveâ genes, encoding enzymes involved in the glyoxylate and Calvin-Benson cycles, gluconeogenesis, and the pentose phosphate pathway, is responsive to culture conditions and genetic background as well as to boosting. Four genesâencoding a diacylglycerol acyltransferase (DGTT2), a glycerol-3-P dehydrogenase (GPD3), and two candidate lipases (Cre03.g155250 and Cre17.g735600)âare selectively upregulated in the sta6 strain. Although the bulk rate of acetate depletion from the medium is not boost enhanced, three candidate acetate permease-encoding genes in the GPR1/FUN34/YaaH superfamily are boost upregulated, and 13 of the âsensitiveâ genes are strongly responsive to the cell's acetate status. A cohort of 64 autophagy-related genes is downregulated by the boost. Our results indicate that the boost serves both to avert an autophagy program and to prolong the operation of key pathways that shuttle carbon from acetate into storage lipid, the combined outcome being enhanced TAG accumulation, notably in the sta6 strain
Genome-wide histone acetylation data improve prediction of mammalian transcription factor binding sites
Motivation: Histone acetylation (HAc) is associated with open chromatin, and HAc has been shown to facilitate transcription factor (TF) binding in mammalian cells. In the innate immune system context, epigenetic studies strongly implicate HAc in the transcriptional response of activated macrophages. We hypothesized that using data from large-scale sequencing of a HAc chromatin immunoprecipitation assay (ChIP-Seq) would improve the performance of computational prediction of binding locations of TFs mediating the response to a signaling event, namely, macrophage activation
MPLEx: a Robust and Universal Protocol for Single-Sample Integrative Proteomic, Metabolomic, and Lipidomic Analyses
ABSTRACT Integrative multi-omics analyses can empower more effective investigation and complete understanding of complex biological systems. Despite recent advances in a range of omics analyses, multi-omic measurements of the same sample are still challenging and current methods have not been well evaluated in terms of reproducibility and broad applicability. Here we adapted a solvent-based method, widely applied for extracting lipids and metabolites, to add proteomics to mass spectrometry-based multi-omics measurements. The m etabolite, p rotein, and l ipid ex traction (MPLEx) protocol proved to be robust and applicable to a diverse set of sample types, including cell cultures, microbial communities, and tissues. To illustrate the utility of this protocol, an integrative multi-omics analysis was performed using a lung epithelial cell line infected with Middle East respiratory syndrome coronavirus, which showed the impact of this virus on the host glycolytic pathway and also suggested a role for lipids during infection. The MPLEx method is a simple, fast, and robust protocol that can be applied for integrative multi-omic measurements from diverse sample types (e.g., environmental, in vitro , and clinical). IMPORTANCE In systems biology studies, the integration of multiple omics measurements (i.e., genomics, transcriptomics, proteomics, metabolomics, and lipidomics) has been shown to provide a more complete and informative view of biological pathways. Thus, the prospect of extracting different types of molecules (e.g., DNAs, RNAs, proteins, and metabolites) and performing multiple omics measurements on single samples is very attractive, but such studies are challenging due to the fact that the extraction conditions differ according to the molecule type. Here, we adapted an organic solvent-based extraction method that demonstrated broad applicability and robustness, which enabled comprehensive proteomics, metabolomics, and lipidomics analyses from the same sample
